INCREASED MITOCHONDRIAL DNA CONTENT IS ASSOCIATED WITH DELAYED EMBRYO DEVELOPMENT AND ANEUPLOIDY

To determine the relationship between mitochondrial DNA content (mtDNA) as reflected by MitoScore to ploidy and embryo morphokinetics (MK) using time-lapse imaging. Patients undergoing in vitro fertilization (IVF) with preimplantation genetic testing for aneuploidy (PGT-A) from January 2020 February 2021 were studied retrospective review of prospectively collected data. MK parameters including time 4 cells (t4), t5, t8, t9, start compaction (tSC), morula (tM), blastulation (tSB), blastocyst (tB) expanded blast (tEB) assessed. Second (cc2) third (cc3) cell cycles duration synchronous division (s2) calculated. Blastocysts graded follows: maturity (1-early, 2-full, 3-expanded, 4-hatched), inner mass (ICM) morphology (1-good, 2-fair, 3-poor/absent) trophectoderm (TE) grade 3-poor/absent). Associations MitoScore, parameters, ploidy, evaluated. Kruskal-Wallis test Spearman correlation performed. A p-value <0.05 was considered statistically significant. Embryos 78 IVF cultured blastocyst. Of 414 blasts biopsied PGT-A analysis, 192 on day 5 202 6. these, 162 embryos diagnosed euploid 223 aneuploid. significant difference seen those v. 6 (20.7 30.2; p<0.001).
